Fig. 4. Older rats expressed <t>less</t> <t>synaptic</t> proteins (arbitrary units) in the hippocampus compared to young rats and the difference is more drastic in low-capacity Runner (LCR) rats than in high- capacity runner (HCR) rats. Western blot analyses of the expression of (A) synapsin (SYN-1) and (B) synaptophysin <t>(SYP)</t> in hippocampal homogenates of HCR and LCR rats at the age of 8 weeks (young) and 10 months (old). The membrane was cut horizontally to separate SYN-1 and SYP according to their molec ular weights, which are shown with the representative blot image. The graphs show the amount of the pro teins normalized to the total protein as analyzed from the stain-free blot. From the stain-free blot only the band of approximately 50 kDa is shown. The boxes and whiskers are the same as in Fig. 1. Mann-Whitney U test. * p ≤0.050, ** p ≤0.010 and *** p ≤0.001.

Article Snippet: Immunostaining was performed using monoclonal antibodies against c‐Fos (1:1000; Cell Signaling Technology, cat#2250S) and synaptophysin (SYP, 1:400; Santa Cruz, sc‐17750), polyclonal antibody against PSD95 (1:200; Invitrogen, cat#51‐6900), followed by incubation at 4°C for 24 h. Goat anti‐ mouse (Alexa Fluor 594, 1:500, ab150116) and goat anti‐rabbit (Alexa Fluor 488, 1:500, ab150077) IgG were used as secondary antibodies and incubated in the dark at room temperature for 2 h. Fluorescent images were captured using an ECHO Revlove fluorescence microscope (ECHO, San Diego, California, USA) and quantitatively analyzed using ImageJ software (NIH, Bethesda, Maryland, USA).
